Il film è ispirato al libro dello stesso anno di Nicholas Pileggi Casino: Love and Honor in Las Vegas, che dettaglia le attività criminali dei mafiosi statunitensi Frank "Lefty" Rosenthal e Anthony "The Ant" Spilotro nel mondo dei casinò della Las Vegas degli anni ottanta. Viene spesso considerato la terza parte di una trilogia del regista sulla mafia, iniziata nel 1973 con Mean Streets - Domenica in chiesa, lunedì all'inferno e proseguita nel 1990 con Quei bravi ragazzi.
Il film segue De Niro, un giocatore d'azzardo ebreo-americano esperto, incaricato dalla mafia di Chicago di supervisionare le operazioni quotidiane al Tangiers Casino di Las Vegas. I personaggi includono Joe Pesci e Sharon Stone, una truffatrice di strada che il protagonista sposa e con cui ha una figlia. Il film descrive in dettaglio la gestione del casinò da parte di De Niro, le difficoltà che affronta nel suo lavoro, il coinvolgimento della mafia e il graduale crollo delle sue relazioni, con Las Vegas che cambia sullo sfondo.
Negli Stati Uniti, il film uscì il 22 novembre 1995, arrivando in Italia il 22 marzo 1996.
Per la sua interpretazione, la Stone venne candidata all'Oscar e vinse il Golden Globe per la migliore attrice in un film drammatico, mentre Scorsese ottenne una candidatura a quello per il miglior regista.
